i'm gonna throw The Cannonball Run (1981) out there, 'cause it never ceases to make me very happy. and i just this morning re-watched The Goonies (1985) for the 500th time. just a whole lotta love there, too...

Can I have two? Four Rooms (1995) not only has a great ensemble cast (Tim Roth, Antonio Banderas, Madonna, Bruce Willis, Maris Tomei and Quentin Tarantino) to name a few. It also has an ensemble group of directors; Allison Anders, Alexandre Rockwell, Robert Rodriguez and Quentin Tarantino, it's a great film if you haven't checked it out.
And the other is CONfidence (2003), a con flick starring Ed Burns, Rachel Weisz, Andy Garcia, Dustin Hoffman and Paul Giamatti.

It is not so much an ensemble cast as much as a boat-load of cameos, but the hilarious comedy Run Ronnie Run (2002) from David Cross and Bob Odenkirk is one of my favs.

Ensemble casts: Magnolia (1999), Tombstone (1993), Heat (1995), LA Confidential (1997) and Usual Suspects (1995). I consider Ocean's (2001) exempt for being totally obvious.

Parenthood (1989): Jason Robards was so good in that film. Crash (2004): Thandie Newton freaking out when Matt Dillon has to save her...*shivers*.

Either The Player (1992) (more cameos then cast), or A Few Good Men (1992) - Cruise, Moore, Nicholson, Kevin Bacon, Noah Wylie, Cuba Gooding, Jr, Keifer Sutherland, Christopher Guest, and the immortal, awesome, JT Walsh. Even Aaron Sorkin has a silent role as a bar patron. Now that's a cast!

Probably Short Cuts (1993), it any other altman movie, he knew how to get stars as extras, though the expendables might beat that when out.

I really liked Galaxy Quest (1999) in terms of ensemble. Was that a movie people missed? I can't recall seeing it in theaters..

My favorite ensemble cast is undoubtedly Kurosawa's Seven Samurai (1954). Everyone is perfect, and dare I say Magnificent....GET IT!? ;)

My first thought was Gosford Park (2001).  That movie kinda blew my mind as each actor made their entrance.

Normally the trick for me is to find a film that no one else has suggested, but surprisingly no one mentioned any of the Christopher Guest movies, so I claim them!  They are all mine.  mwah haha.
