i would go with Sam Rockwell in Moon (2009). it was not just surprising that Rockwell was the lead, but also that he carried the film as more or less the sole character. thanks again MYMHM for calling this film to my attention.

Maybe, Jim Carrey in Eternal Sunshine of the Spotless Mind (2004)? I was used to just seeing him as a guffy shmuck before that movie. Similarly but opposite, Joseph Gordon-Levitt in Brick (2005) was the first time I saw 'that kid from 3rd rock from the sun' in a serious movie.

I've said it before and I'll say it again.  David Warner in Teenage Mutant Ninja Turtles 2: Secret of the Ooze (1991). How the eff did they get him?

I know it's not technically a movie.  More of a Mini-series.  But I was initially surprised to see David Tennant as the romantic lead in Viva Blackpool (2004).  But by the end, I totally bought it.  He won me over big time.
